Transplanted human embryonic stem cells as biological 'catalysts' for tissue repair and regeneration.

Human embryonic stem cells have tremendous potential in the newly emerging field of regenerative medicine. Recently, it was demonstrated that the rescue of lethal cardiac defects in Id knockout mutant mouse embryos was not due to the transplanted cells giving rise to functional new tissues within the defective embryonic heart. Instead, there is indirect evidence that the observed therapeutic effect was due to various secreted factors emanating from the transplanted cells. This therefore, introduces the exciting prospect of utilizing human embryonic stem cells as biological 'catalysts' to promote tissue repair and regeneration in transplantation therapy. However, the immunological barrier against allogenic transplantation, as well as the teratogenic potential of human embryonic stem cells poses major technical challenges. A possible strategy to overcome the immunological barrier may be to impose a temporary regimen of immunosuppressive drugs followed by their gradual withdrawal, once adequate tissue regeneration has been achieved. Other more novel alternatives include the use of microencapsulation to block interaction with the transplant recipient's immune system, and co-transplantation with bone marrow-derived mesenchymal stem cells, which have been demonstrated to possess immuno-suppressive properties. The teratogenic potential of human embryonic stem cells could possibly be alleviated by directing the differentiation of these cells to specific lineages prior to transplantation, or through mitotic inactivation (gamma irradiation or mitomycin C exposure). Co-transplantation with autologous adult stem cells may represent a novel strategy to further enhance the 'catalytic' effects of human embryonic stem cells. The various factors secreted by human embryonic stem cells could then have a concentrated localized effect on relatively large numbers of co-transplanted autologous adult stem cells, which may in turn lead to enhanced repair and regeneration of the damaged tissue or organ. Moreover, there is also a possibility that synergistic interactions between the co-transplanted human embryonic stem cells and autologous adult stem cells, may somehow produce signals for the recruitment and migration of additional endogenous adult stem cells within the recipient (i.e. peripheral blood circulation, bone marrow), which could further enhance organ/tissue regeneration. Hence, the potential use of human embryonic stem cells as biological 'catalysts' to stimulate tissue repair and regeneration, appears to hold tremendous promise in the field of regenerative medicine. This new therapeutic strategy needs to thoroughly investigated, in view of its potentially important clinical applications.

최근 ‘냉동제대혈로부터 줄기세포를 분리 및 배양하는 방법’에 대한 원천 기술이 미국 특허청에 등록된 이후 연이은 쾌거라고 밝혔다.
이번 특허내용은 만삭의 성숙한 태아에서 획득한 제대혈에서 중간엽 줄기세포를 추출 및 배양하는 방법에 대한 원천기술로서 거의 모든 산모로부터 출산한 제대혈이 이에 해당한다고 업계는 전하다.
Research Impact Technologies(2008년 5월) 보고서에 따르면 유럽 줄기세포 시장은 2009년 4,789 백만달러에서 2012년 9,389 백만달러로 연평균 26.58%로 성장하고 있는 세계적 파급효과가 큰 산업이다. 또한, 이번 유럽 특허 등록은 유럽의 27 개국의 모든 나라에서 특허 권리를 20년간 행사할 수 있다.
㈜히스토스템은 이번 미국에 이은 유럽 특허등록은 제대혈 줄기세포 획득 원천기술이 상업화되면 세계 줄기세포시장이 크게 활성화되고 우리나라가 세계시장의 중심에 설 수 있을 것으로 기대하고 있다. 또한 이 원천기술을 이용하면 제대혈에서 줄기세포를 대량 공급하는 것이 가능해져 전 세계 난치성 질환을 앓고 있는 환자들에 대한 제대혈 줄기세포 공급 인프라가 구축될 수 있을 것으로 내다보고 있다

제대혈줄기세포 전문기업 ㈜히스토스템과 김휘율 건국대 수의과대 교수는 "세계 최초로 사람 제대혈로부터 분리한 탯줄 줄기세포를 뇌졸중 개에 이식, 치료에 성공했다"고 밝혔다. 김휘율 교수는 "히스토스템이 공급한 탯줄혈액 줄기세포로 뇌졸중을 유발한 비글견에 뇌바닥 동맥 내로 줄기세포를 주입한 결과 손상 받은 뇌병변에서 탯줄혈액 줄기세포가 존재하고 있었으며 줄기세포의 신경세포로 분화도 확인했다"고 말했다. 특히 "뇌바닥 동맥 내로 줄기세포를 주입해 뇌졸중을 치료한 첫 케이스로 뇌병변 부위에 직접 주입하지 않고 비침습적 치료방법을 개발함으로서 향후 줄기세포를 이용한 뇌졸중 치료에 있어 큰 방향성을 제시했다"고 지적했다. 사람의 뇌졸중 치료에 탯줄혈액 줄기세포가 중요한 역할을 한다는 것과 줄기세포를 뇌병변에 직접 주사하는 방법을 바꾸는 새로운 줄기세포 치료방법을 제시한 이번 연구결과는 국제 신경과학전문 학술지 <저널오브뉴로사이언스리서치(Journal of Neuroscience Research)>에 최근에 게재됐다. |

| 3 main types of stem cells are being used: • Neural Stem Cells (derived from fetal stem cells) used for: Cerebral Palsy, Brain Injury, Stroke, Degenerative Diseases, and other neurological disorders. • hRPE Stem Cells (Adult Retinal Stem Cells) Used for: Parkinson's Disease • Bone Marrow Stem cells (Adult Bone Marrow stem cells extracted from the patient's own bone marrow) Recommended for young patients with a weak immune system. 좀 이상하지 않나요? fetal stem cell이라는 것은 넓은 의미로 태아 자체의 세포와 제대혈을 포함하는 용어인데, 제대혈중의 HSC는 신경세포가 되지 못하므로, 제대혈중의 MSC를 사용하거나 태아 자체의 세포를 쓴다는 이야기가 되는데요. 제대혈중의 MSC는 그 양이 너무 적어서 사용이 곤란다하고 하는데. 그럼 정말 태아자체의 세포를 체취해서 쓴다는 이야기인가????? 두번째의 경우도 , 홈피를 잘 읽어보면, single donor 에서 얻는다고 하는데 그럼 멀쩡한 사람의 눈을 찌른다는 이야긴가??? 중국에서 장기밀매가 공공연한 비밀이라는 것은 다 아는 사실인데 정말 이게 사실이라면, 중국에서 줄기세포치료가 발전하는 이유도 알겠네요. 이렇게 태아를 죽이거나 멀쩡한 사람 눈을 빼는 것 말고는 방법이 없을까요? 척수손상의 경우에, 배아줄기세포와 성체줄기세포를 혼합하여 주입을 하여 성공한 경우가 있습니다. 뭐 , 이경우에도 태아대신 배아가 필요할 뿐이므로, 윤리적인 문제를 완전히 벗어날 순 없네요. 배아대신, IPS가 상용화되길 기다려 봐야 겠습니다. | ||

In May 2001, Israeli parents of a nine-year old boy with a crippling disease that left him wheelchair-bound took their child to see doctors in Moscow. In a highly experimental procedure that was presumably unavailable in their home country, those doctors injected fetal stem cells into various regions of his brain.
The boy’s parents—they aren’t named in a report describing the case in this week’s PLoS Medicine—must have been desperate. The nine-year old suffered from ataxia-telangiectasia, a childhood disease that causes degeneration of parts of the brain that control muscle movements and speech. The symptoms include slurred speech, poor balance, impaired immune function, and the appearance of red spider veins called telangiectasias in the eyes, ears or cheeks.
There are no treatments for the disorder and the prognosis is dim; patients usually only make it into their teens or early twenties, according to the National Institute of Neurological Disorders and Stroke. While it’s unclear exactly what the Russian doctors were trying to achieve (the researchers who wrote the case report were not involved in the stem cell therapy), they must have been hoping that the injected cells would restore some function in his brain, or at least slow the disease progression. The boy went back for injections in 2002 and 2004, although it’s not clear from the report whether his condition improved as a result.
Then he was diagnosed with a brain tumor in 2005. That tumor, it turns out, grew out of the stem cells, obtained from at least two aborted fetuses, used in his brain.
이 러시아 의사는 pleuripotent 한 fetal stem cell이 주위 환경이 brain 신경들이므로,
주위 영향을 받아 신경으로 분화할 것이라 기대를 한 모양입니다.
그 기대를 어기고, 이넘은 종양이 되었네요.
워낙 능력이 좋아서 주위 환경의 영향을 별로 안 받는 놈이네요.
MSC의 경우에는 주위의 영향을 받는다고 하는데, 역시 ES나 fetal stem cell은
pleuripotent하군요.
어쨌든, 무식하고 용감했던 러시아의사 덕에 아이는 종양이 생겨버렸습니다.
이경우, adult stem cell과 mix해서 넣었더라면 종양이 안생겼을 수도 있지요.
실제, spinal cord에 넣어서 종양이 안생기고 마비를 회복한 경우가 있지요.
또는, neural stem cell로 1차 분화를 시켜서 넣는 것도 한 방법인데,
이경우에도 안전하게 할려면, BM derived adult stem cell을 같이 mix하는게
좋을 것 같습니다.
